ABSTRACT

Screening and preventive treatment for latent tuberculosis infection (LTBI) are important measures to reduce the incidence of active tuberculosis (TB) and its global burden. This paper discusses the current status of LTBI prevalence and preventive treatment, risk of incidence in high-risk groups, screening methods, and other interventions, emphasizing the importance of prevention and control as early as possible in a scientific strategy, so as to lay a solid foundation for eliminating TB by 2030.

ABSTRACT

Objective To systemically analyze family burden, quality of life of chronic hepatitis B and C patients in Shanghai and related influencing factors. Methods A representative sample of chronic hepatitis patients (n=1 478) and their family members (n=1 478) was randomly selected through a multi-stage cluster sampling from 30 communities in 10 districts of Shanghai. One patient and one family member of each family were interviewed using different questionnaires to collect related information.Based on Bronfenbrenner's ecological systems, psychological measurement,two-level random intercept model and multivariable structural equation model were applied to determine the effects and directions of the factors between life quality of chronic hepatitis patients and family burden. Results The mean score of quality of life of chronic hepatitis patients in Shanghai was 78.70 ± 13.25,the score of "specific module" was highest and the score of "social function" was lowest.Additionally, the mean score of burden reported by the family members was 12.62 ± 10.74,the score of"financial burden" was highest, and the score of"effect on family member's health" was lowest.Multivariable structural equation model indicated that eight factors were related with life quality and family burden of patients with chronic hepatitis.Among them,HCV infection, elevated serum alanine aminotransferase level, average monthly cost for patient >3 000 yuan (RMB) and poor health of family members were the direct risk factors for the life quality of the patients as well as family burden. The factor of drinking more than once a week influenced the patients' life quality directly and family burden indirectly. On the contrary, the factors of local household registration, hospitalization and family member's indifferent attitude to hepatitis B vaccination influenced the family burden of the chronic hepatitis patients directly and the life quality of the patients indirectly. Conclusion The findings could be used in the development of community based management and intervention of chronic hepatitis patients in Shanghai.

ABSTRACT

Objective To investigate the molecular epidemiology of human immunodefieiency viruses (HIV)-1 infected individuals in Honghe district,Yunnan Province and provide the evidence of molecular biology features of HIV-1 infection. Methods HIV-1 pol gene was amplified by reverse transcription-polymerase chain reaction (RT-PCR).Then sequencing and phylogenetic tree analysis were employed tO determine HIV-I subgenotype. The sequence alignment was performed in the database of international drug resistance tO identify resistance-associated mutations. Results The samples from 60 HIV-1 infected individuals were investigated:39 were male,21 were female, with average age 35.5 years old.Thirty-four cases were infected with HIV-I through intravenous drug abuse,12 by sexual contacts,2 were contaminated blood/blood products transfusion and 12 with unknown transmission routes.Phylogenetic analysis revealed that 53 cases (88.3%) were subtype 08-BC,6 (10.0%) were subtype 07-BC and 1 (1.7%) was subtype 01_AE.The total rate of drug resistance associated mutations was 33.3%.The major mutations in protease (PR) and reverse transcriptase (RT) regions accounted for 5.0% and 3 1.7%,respectively.The major mutations in PR region were I541M,V82VFIL,M46MI,which were found in 1 case, respectively. The mutations in RT region were as follows:4 cases were T69D,6 were A62V,1 was D67DE,1 was E44D,3 were V179D,1 was V179E.1 was K238KN,1 was L234T+P236S and 1 was V106E.Conclusions The major transmission route of HIV-I infection in Honghe district,Yunnan Province is through drug injection. The major HIV-1 subtype of HIV-infeeted individuals is 08_BC.PR inhibitor and RT inhibitor drug resistance associated mutations in HlV-1 gene have already existed.

ABSTRACT

Objective:To investigate the impact of the CD4+CD25nt/hiCD127lo regulatory T cell subset frequency on the immune status and disease progression of Chinese HIV-1 infected individuals.Methods:83 untreated HIV-infected individuals and 312 healthy control individuals of four distinct age groups were enrolled in the research. The CD4+ T cell absolute counts, phenotypes and frequency determination of CD4+CD25nt/hiCD127lo Regulatory T cell subsets was performed on freshly obtained whole blood samples by 3-color immune staining flow cytometry. The HIV-1 specific cellular immune function was test at single cell level by ELISpot. The corresponding plasma viral load was determined by NASBA.Results:The frequency of peripheral CD4+CD25nt/hiCD127lo regulatory T cells of HIV infected individuals in distinct disease progression status was dissimilar in China , and significantly increased in contrast to the healthy controls(P

ABSTRACT

The effect of CHIP [cis-dichloro-trans-dihydroxy-bis-isopropy-lamine platinum (Ⅳ)] on the biosynthesis of DNA, RNA, and pro-tein of Ehrilch ascites carcinoma ( EAC ) cells in vitro was studied. 3H-TdR, 3H-UR, 3H-Leu were used as precursors of cellular macro-molecules to study the effect of CHIP on the biosynthesis, with CDDP [cis-dichlorodiammineplatitinum (Ⅱ)] as a positive control. The experimental results showed that CHIP, similar to CDDP, inhibited the biosynthesis of DNA, RNA and protein, and has a strong antitumor activity. The inhibitory effect on DNA biosynthesis lasted 40 hours indicating that CHIP might induce permanent damage in DNA template and thus reduced the mitotic division of tumor cells. Furthermore, in contrast to the general view, the in vitro experimental results showed that CHIP, a tetravalent platinum coordination compound, exerted its antitumor activity directly in vitro.
